What exactly is bromantane?

Bromantane (ADK-709, trade name Ladasten) is N-(4-bromophenyl)adamantan-2-amine — an adamantane-derived secondary amine of the 2-aminoadamantane family, developed in the 1980s at the Zakusov Institute of Pharmacology in Moscow. It is classified as an actoprotector: a synthetic adaptogen that enhances stability against physical loads without increasing oxygen consumption or heat production.

Is bromantane a stimulant?

No. Psychostimulants force monoamines out of their stores and leave the system overdrawn; bromantane instead induces expression of tyrosine hydroxylase and DOPA-decarboxylase, the enzymes that synthesise dopamine from scratch. The actoprotector profile is one of non-exhaustive action: no hyperstimulation during the effect, no crash after it, and no withdrawal on discontinuation in the clinical trials.

Is bromantane a urea derivative?

No — that is a common error in marketing copy. The verified structure (PubChem CID 4660557) is an adamantane-derived secondary amine: an adamantane cage joined through nitrogen to a para-bromophenyl ring, C₁₆H₂₀BrN, 306.24 g/mol. There is no urea moiety in the molecule.

How does bromantane work?

Its best-documented action is genomic: it upregulates the genes for tyrosine hydroxylase and DOPA-decarboxylase in dopamine-producing brain regions, raising de novo dopamine synthesis. An epigenetic correlate — demethylation of the TH gene promoter — has been reported. It is not a meaningful dopamine reuptake inhibitor (that requires 50–500 µM in vitro), and the receptor or target upstream of the gene-expression effect remains unknown. Was bromantane used by cosmonauts?

No source supports that claim. The space-programme actoprotector was bemitil (Metaprot) — the first recipients of bemitil were Soviet cosmonauts. The cosmonaut story attached to bromantane is a marketing embellishment that conflates the two compounds, and this monograph does not repeat it.

What is bromantane's regulatory status?

In Russia bromantane is an approved prescription medicine (Ladasten, 50 mg tablets) for asthenic disorders, in the State Register of Medicines. In the United States it is unscheduled but not FDA-approved. No EU or UK marketing authorisation has been identified, so elsewhere it is treated as a research compound. Laws differ by country; check your own jurisdiction.

Is bromantane banned in sport?

Yes. After several Russian athletes tested positive at the 1996 Atlanta Olympics — the detection was reported in The Lancet in 1997 — bromantane was prohibited in sport from 1997. Its metabolites remain detectable in urine for two weeks or more, which keeps it a doping-control target. Athletes should consult the current WADA Prohibited List.

Does bromantane cause dependence or withdrawal?

The verified record says no. The placebo-controlled neurasthenia trial found no withdrawal syndrome after discontinuation, and chronic animal administration showed no dependence or tolerance. In the 728-patient trial, adverse effects occurred in 3% of patients with no serious adverse events at 50–100 mg/day over 28 days.
